About Matrixyl

Palmitoyl pentapeptide-4 is the five-amino-acid sequence Lys-Thr-Thr-Lys-Ser (KTTKS) with palmitic acid, a 16-carbon fatty acid, attached to one end. KTTKS is a fragment of the C-terminal propeptide of type I procollagen, and the fatty acid is there to help it get through the outer layer of skin.

It was launched as Matrixyl by the French cosmetic ingredient company Sederma in 2000, and it’s still one of the best-known signal peptides in skin care research.

What studies report about Matrixyl

In 1993 Katayama and colleagues reported in the Journal of Biological Chemistry that KTTKS stimulated fibroblasts to produce more type I and type III collagen and fibronectin in culture.

A 12-week, vehicle-controlled study of palmitoyl pentapeptide-4 on facial skin, published by Robinson and colleagues in 2005 in the International Journal of Cosmetic Science, reported improvements in fine lines and wrinkles measured by image analysis.

The full list of Matrixyl papers is on PubMed, and most abstracts are free to read.

How Matrixyl works

Collagen fragments made when old collagen breaks down are thought to act as a feedback signal: they tell fibroblasts that the matrix needs repair. KTTKS mimics one of those fragments.

In fibroblast cultures that signal shows up as more collagen, fibronectin and glycosaminoglycan production. The palmitoyl group makes the peptide more fat-soluble, which matters for how well it crosses skin in topical work.

Using Matrixyl 200mg in formulation work

Scoop or weigh the powder quickly and reseal the pouch or jar. Humidity is the main enemy of a dry peptide, more than heat.

Most cosmetic peptides dissolve in water or in the water phase of a formula. Add them below about 40°C, after the hot steps, so heat doesn’t damage the chain.

Keep the unopened powder frozen. Once you’re working from it, the fridge and a tight seal are enough for a month or so.

Study designs that use Matrixyl 200mg

A typical path starts with cell culture: fibroblasts for matrix proteins, melanocytes for pigment, nerve and muscle co-cultures for expression-line peptides. Reconstructed skin equivalents come after that.

Run a vehicle-only batch beside every peptide batch. Without a control made from the same base, it’s hard to say whether a change came from the peptide or from the cream.
