PE-22-28 at a glance
PE-22-28 is a seven-amino-acid peptide, Gly-Val-Ser-Trp-Gly-Leu-Arg, shortened from spadin. Spadin comes from the propeptide of sortilin, a protein involved in moving other proteins around inside neurons.
It was developed by Jean Mazella’s group in France as a more stable version of spadin. It’s a research compound only.
What studies report about PE-22-28
Mazella and colleagues described spadin in PLoS Biology in 2010 as a blocker of the TREK-1 potassium channel with antidepressant-like effects in mice.
In 2017, Djillani and colleagues reported shorter analogs, including PE-22-28, that lasted longer and showed similar effects in mouse models within four days.
For the primary literature, PubMed lists the PE-22-28 studies.
PE-22-28 mechanism of action
PE-22-28 blocks TREK-1, a potassium channel that keeps neurons quiet. Mice lacking TREK-1 are resistant to depression-like behavior in standard tests, which is what made the channel a target.
In the mouse studies, blocking it also raised markers of new neuron formation in the hippocampus.

How to mix PE-22-28 8mg
The concentration is the 8mg in the vial divided by the water you add:
- 1mL of water: 8mg per mL
- 2mL of water: 4mg per mL
- 3mL of water: 2.67mg per mL
Let the vial warm to room temperature first. Run the water slowly down the side of the glass, then swirl or roll it until the powder is gone. Don’t shake it; foam is a sign of damaged peptide.
Dry vials last longest frozen at -20°C. Reconstituted solution goes in the fridge and is best used within four weeks or so.
Planning a study with PE-22-28 8mg
Work out the total amount the protocol needs first, then add about 20% for test runs and the odd lost dose. Ordering everything from one lot keeps the material consistent through the whole study.
Where PE-22-28 fits
PE-22-28 is a short spadin analog that blocks the TREK-1 potassium channel. If you’re choosing between options, these are the usual alternatives:
- Semax is built on a fragment of ACTH and linked to BDNF in rodent studies
- N-Acetyl Semax is Semax with protective groups on both ends so it lasts longer in solution
- Selank is built on tuftsin and studied for anxiety through the GABA system
- N-Acetyl Selank is Selank with protective groups on both ends
- DSIP is a nine-amino-acid peptide first isolated from rabbits in deep sleep
These reach the brain through different systems: BDNF, GABA, sleep regulation and a potassium channel. All of them are on our nootropic peptides page.
Related nootropic peptides
- Selank 10mg, a tuftsin analog
- DSIP 5mg, a neuropeptide
