What is AICAR?

AICAR (5-aminoimidazole-4-carboxamide ribonucleoside, also called acadesine) is a small molecule, not a peptide. Inside the cell it’s phosphorylated to ZMP, which looks enough like AMP to switch on AMP-activated protein kinase (AMPK), the cell’s energy sensor.

It’s been a standard lab tool for activating AMPK since the 1990s. Acadesine was also tested in heart surgery trials as a way to protect heart muscle from ischemia, but it never reached approval.

What studies report about AICAR

A 2008 paper in Cell by Narkar and colleagues at the Salk Institute reported that four weeks of AICAR improved running endurance in sedentary mice and shifted muscle gene expression toward an endurance profile, which is why it gets called an exercise mimetic. The World Anti-Doping Agency banned it the following year.

In metabolic research AICAR is used to raise glucose uptake in muscle, to increase fatty acid oxidation and to study how AMPK signaling interacts with insulin pathways in cell and rodent models.

How AICAR works

AMPK switches on when the cell’s energy charge falls, meaning the ratio of AMP to ATP goes up. Once active it turns on pathways that make ATP, such as glucose uptake and fat burning, and turns off ones that use it, such as fat and cholesterol synthesis.

ZMP binds the same site on AMPK that AMP does, so AICAR activates the enzyme without the cell actually being short of energy. That’s what makes it useful for separating the effects of AMPK from the effects of exercise or fasting.

Reconstituting AICAR 50mg

Let the vial warm to room temperature first. Run the water slowly down the side of the glass, then swirl or roll it until the powder is gone. Don’t shake it; foam is a sign of damaged peptide.
