PT-141, also called bremelanotide, is a cyclic seven-amino-acid peptide developed from Melanotan II. It activates melanocortin receptors in the brain, mainly MC4R.

Palatin Technologies developed it, and the FDA approved it in 2019 as Vyleesi for hypoactive sexual desire disorder in premenopausal women.

PT-141 activates MC4R and MC3R in the hypothalamus, receptors involved in sexual motivation and arousal pathways. It acts on the brain, not on blood flow, which sets it apart from PDE5 inhibitors.

Chemically it’s Melanotan 2 with a free acid where Melanotan 2 has a C-terminal amide.

Divide 10mg by the volume of water and you have the concentration:

• 1mL of water: 10mg per mL
• 2mL of water: 5mg per mL
• 3mL of water: 3.33mg per mL

Bring the vial to room temperature, add sterile bacteriostatic water against the glass and swirl gently. It should go clear within a minute or two. Shaking only makes foam.

Store sealed vials in the freezer. Once you’ve added water, keep the vial at 2 to 8°C and plan to finish it within roughly a month.
