About NAD+

The compound (nicotinamide adenine dinucleotide) is a coenzyme found in every living cell. It carries electrons in the reactions that turn food into ATP, and it’s used up by enzymes such as the sirtuins, PARPs and CD38.

Levels fall with age in several tissues in rodents and people, which is why the compound and its precursors, such as NMN and nicotinamide riboside, have become a major topic in aging research.

NAD+ research so far

Work from Shin-ichiro Imai’s lab at Washington University linked falling the compound to age-related decline in mice. In a small human trial published in Science in 2021, Yoshino and colleagues reported that the precursor NMN improved muscle insulin sensitivity in prediabetic women.

Most studies raise the compound with precursors such as NMN or nicotinamide riboside. Direct the compound work is a separate research question.

How NAD+ works

The compound shuttles electrons in glycolysis, the Krebs cycle and the mitochondrial chain, so it’s needed to turn food into ATP. It’s also used up by sirtuins, which regulate gene expression, and by PARPs, which repair DNA.

Because those enzymes consume it, the compound levels depend on constant recycling. Falling levels with age are linked to lower sirtuin activity, which is the basis for most the compound aging research.

NAD+ reconstitution and storage

For the 100mg size, here’s the concentration you get with common volumes of bacteriostatic water:

• 2mL of water: 50mg per mL
• 3mL of water: 33.33mg per mL
• 5mL of water: 20mg per mL

Use sterile bacteriostatic water and add it slowly, aiming at the glass instead of the powder. A gentle swirl is all it needs.

Before mixing, keep it at -20°C and out of the light. After mixing, the fridge at 2 to 8°C, and use it within about four weeks.
