What is LL-37?

LL-37 is the only cathelicidin antimicrobial peptide humans make. It’s 37 amino acids long and starts with two leucines, which is where the name comes from. It’s cut from a larger precursor protein, hCAP-18, and is found in white blood cells, skin and the lining of the airways and gut.

It’s amphipathic, with one charged face and one oily face, so it can sit in and disrupt bacterial membranes.

LL-37 in published research

Lab work shows it kills a wide range of bacteria and some fungi and viruses in culture, and it also acts as a signaling molecule in inflammation and wound repair. Heilborn and colleagues reported in 2003 that it’s needed for normal re-epithelialization of human skin wounds in culture.

Human data are limited to small early-phase studies, for example on venous leg ulcers.

For the primary literature, PubMed lists the LL-37 studies.

The biology behind LL-37

LL-37 has a positively charged face that’s drawn to the negatively charged membranes of bacteria. Once it’s there, its oily face pushes into the membrane and makes holes.

It does more than kill microbes. It binds bacterial endotoxin, recruits immune cells and stimulates new blood vessel growth, which ties it into wound healing studies.

LL-37 5mg reconstitution and storage

Divide 5mg by the volume of water and you have the concentration:

  • 1mL of water: 5mg per mL
  • 2mL of water: 2.5mg per mL
  • 3mL of water: 1.67mg per mL

Use sterile bacteriostatic water and add it slowly, aiming at the glass instead of the powder. A gentle swirl is all it needs.

Dry vials last longest frozen at -20°C. Reconstituted solution goes in the fridge and is best used within four weeks or so.

Planning a study with LL-37 5mg

Map out the doses per group, multiply by the number of days, and add a margin. It’s cheaper to order one extra vial at the start than to restart a study.

Where LL-37 fits

In short, LL-37 is the only human cathelicidin, an antimicrobial peptide that also signals in wound repair. Here’s how its neighbors differ:

  • Thymosin Alpha-1 is a 28-amino-acid thymic peptide, approved in several countries as thymalfasin
  • VIP is a 28-amino-acid neuropeptide that relaxes smooth muscle and calms immune cells
  • Thymalin is a thymus peptide complex from the Leningrad bioregulator program
  • PNC-27 is a p53-derived peptide that binds HDM-2 on cancer cell membranes

All are immune research tools, but they act at very different points, from T cell maturation to direct antimicrobial action. All of them are on our immune peptides page.
