About PNC-27

PNC-27 is a 32-amino-acid peptide made of two parts: residues 12 to 26 of the tumor suppressor protein p53, the stretch that binds HDM-2, joined to a membrane-residency sequence called penetratin. It came from Matthew Pincus and colleagues at SUNY Downstate in New York.

It’s a research peptide only. It hasn’t been through clinical trials and isn’t approved for any use.

PNC-27 research so far

Cell culture papers from the Pincus group report that PNC-27 kills a range of cancer cell lines, including pancreatic, breast and leukemia cells, while leaving normal cells such as fibroblasts and stem cells largely intact.

Their explanation is that many cancer cells carry HDM-2 on their outer membrane. Most of the published work comes from that one group, so independent data is limited.

Search the original papers yourself on PubMed.

PNC-27 mechanism of action

The p53 part of PNC-27 binds HDM-2 on the cancer cell’s membrane. Once bound, the peptide forms pores in the membrane and the cell dies by necrosis, which is much faster than programmed cell death (apoptosis).

Normal cells mostly keep HDM-2 inside, so in culture the peptide has much less to bind on them. That selectivity is the main reason PNC-27 is studied.

Reconstituting PNC-27 5mg

Divide 5mg by the volume of water and you have the concentration:

  • 1mL of water: 5mg per mL
  • 2mL of water: 2.5mg per mL
  • 3mL of water: 1.67mg per mL

Let the vial warm to room temperature first. Run the water slowly down the side of the glass, then swirl or roll it until the powder is gone. Don’t shake it; foam is a sign of damaged peptide.

Dry vials last longest frozen at -20°C. Reconstituted solution goes in the fridge and is best used within four weeks or so.

Ordering enough PNC-27 5mg

Work out the total amount the protocol needs first, then add about 20% for test runs and the odd lost dose. Ordering everything from one lot keeps the material consistent through the whole study.

PNC-27 compared with similar peptides

In short, PNC-27 is a p53-derived peptide that binds HDM-2 on cancer cell membranes. Here’s how its neighbors differ:

  • Thymosin Alpha-1 is a 28-amino-acid thymic peptide, approved in several countries as thymalfasin
  • LL-37 is the only human cathelicidin, an antimicrobial peptide that also signals in wound repair
  • VIP is a 28-amino-acid neuropeptide that relaxes smooth muscle and calms immune cells
  • Thymalin is a thymus peptide complex from the Leningrad bioregulator program

All are immune research tools, but they act at very different points, from T cell maturation to direct antimicrobial action. All of them are on our immune peptides page.
